Purpose of the study: To observe the effects of solasonine and solamargine, alkaloids extracted from Solanum nigrum, on the proliferation of colon cancer cells and explore the possible related signaling pathways. Methods: Three different colon adenocarcinoma cell lines HT-29, HCT116 and SW620, with increasing levels of invasiveness, were used. MTT was used to detect the effect of solasonine and solamargine on cell proliferation; Hoechst 33342 and pSIVA-IANBD / PI staining techniques were used to detect the presence of apoptosis; Western blotting was used to detect the expression of Caspase-3, Caspase-9, Bcl-2, IκB-α, p-IκB-α, p-NFκB P65 in the cytoplasm and NFκB P65 in the nucleus after treatment with solasonine and solamargine; finally, immunocytochemistry was used to observe the effects of the two molecules on NFκB P65 phosphorylation and nuclear translocation in HT-29 cells and HCT116 cells. Results: Both solasonine and solamargine were able, in vitro, to: 1) inhibit the proliferation of HT-29, HCT116 and SW620 cells in a dose-dependent manner; 2) induce apoptosis of HT-29, HCT116 and SW620 cells, with a positive correlation with increasing drug concentration; 3) decrease the expression of p-IκB-α, p-NFκB, increase the expression of IκB-α and modify the expression of Caspase-3, Caspase-9 and Bcl-2; 4) reduce NFκB P65 phosphorylation in HT-29 and HCT116 cells and inhibit NFκB P65 nucleus translocation in the same cells. Conclusions: Both tested molecules were capable, in vitro, to inhibit the proliferation and induce apoptosis of colon cancer cells in a dose-dependent manner and their effect is related to the NFκB pathway.
Scopo dello studio: Osservare gli effetti di solasonina e solamargina, alcaloidi estratti da Solanum nigrum, sulla proliferazione di cellule tumorali del colon ed esplorare le possibili vie di segnalazione correlate. Metodi: Sono state utilizzate 3 differenti linee cellulari di adenocarcinoma del colon HT-29, HCT116 e SW620, che presentano livelli crescenti di invasività. Il test MTT è stato utilizzato per rilevare l'effetto di solasonina e solamargina sulla proliferazione cellulare, le tecniche di colorazione Hoechst 33342 e pSIVA-IANBD / PI sono state utilizzate per rilevare la presenza di apoptosi, il Western blotting è stato utilizzato per rilevare l'espressione di Caspase-3, Caspase-9, Bcl-2, IκB-α, p-IκB-α, p-NFκB P65 nel citoplasma e NFκB P65 nel nucleo dopo il trattamento con solasonina e solamargina; infine, l'immunocitochimica è stata utilizzata per osservare gli effetti delle due molecole sulla fosforilazione di NFκB P65 e sulla traslocazione nucleare nelle cellule HT-29 e nelle cellule HCT116. Risultati: Sia la solasonina che la solamargina sono risultate in grado, in vitro, di: 1) inibire la proliferazione delle cellule HT-29, HCT116 e SW620 in modo dose-dipendente; 2) indurre l'apoptosi delle cellule HT-29, HCT116 e SW620, con una correlazione positiva con l'aumento della concentrazione del farmaco; 3) diminuire l'espressione di p-IκB-α, p-NFκB, aumentare l'espressione di IκB-α e modificare l'espressione di Caspase-3, Caspase-9 e Bcl-2; 4) ridurre la fosforilazione di NFκB P65 nelle cellule HT-29 e HCT116 e inibire la traslocazione del nucleo di NFκB P65 nelle medesime cellule. Conclusioni: Entrambe le molecole testate sono risultate in grado, in vitro, di inibire la proliferazione e indurre l'apoptosi delle cellule del cancro del colon in maniera dose dipendente e il loro effetto è correlato al pathway NFκB.
